What Dosages and Forms Are Spravato Available in?

Spravato is available as a nasal spray.

What is Spravato Used for?

Spravato (Esketamine) is a prescription nasal spray used with oral antidepressants to treat adults with treatment-resistant depression (TRD) and depressive symptoms in adults with major depressive disorder (MDD) with suicidal ideation. The active ingredient in Spravato is Esketamine. Esketamine belongs to the class of medication called antidepressants. It increases the glutamate level, generates new neural paths, and increases brain activity. By this mechanism, there is an increase in the production of neurotransmitters (serotonin and dopamine) in the brain, improving mood and brain activities. Treating depression prevents low mood and feelings of self-harm or suicidal ideation.

Are there any Precautions and Drug Interactions for Spravato?

Before using Spravato nasal spray, tell your doctor if you are allergic to Esketamine, ketamine, or any other ingredients in Spravato. Spravato is a spray and should be used according to your doctor’s instruction. Spravato may cause sleepiness (sedation), fainting, dizziness, spinning sensation, anxiety, or feeling disconnected from yourself, your thoughts, feelings, space, and time. Tell your doctor immediately if you cannot stay awake or will pass out. Your doctor will monitor you for serious side effects for at least 2 hours after taking Spravato, and your doctor will decide if you are ready to leave the hospital. This medication may cause an increase in your blood pressure. You should check your blood pressure regularly and inform your doctor if it is above normal. You should avoid alcoholic beverages while treating depression with Spravato. You should notice an improvement in your condition after four weeks of regular use. However, discuss with your doctor if there is no significant improvement or your symptoms worsen.

You should not take Spravato and inform your doctor about your medical history, especially if you have or have had:
• Asthmatic or other respiratory diseases;
• Diagnosed with arteriovenous malformation;
• Heart diseases such as hypertension, heart attack and heart failure;
• History of aneurysm or intracranial bleeding;
• History of depression/suicidal thoughts;
• Kidney disease;
• Liver disease

Inform your doctor if you are pregnant, plan to become pregnant or breastfeeding a baby. During pregnancy, Spravato may harm the fetus. It would be best to use effective contraception to prevent you from getting pregnant. Esketamine passes into breast milk, and there is a possibility that your baby may be affected. You should not breastfeed during treatment with Spravato.

Tell your doctor about all the medications you use, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements, to prevent drug interactions which might be fatal to your health. This interaction may make your drug less effective, cause adverse effects, or potentiate other drugs' actions. There may be a drug interaction between Spravato and other medications such as:
Cyclobenzaprine;
• Cetirizine
• Hydrocodone;
• Lorazepam;
• Zolpidem
